Lipid-Based Drug Delivery Systems in Vaccines

As a leader in human health improvement, Creative Biolabs has successfully established both liposome development platforms and pre-clinical vaccine platforms. We have absolute advantages in novel drug delivery system services, especially for vaccines.

Background of Vaccines

A vaccine is a special biological preparation administered to healthy individuals with the ability to stimulate immunity and protect the human body from infections and diseases. Vaccines are suspensions of weakened or killed microbes, fragmented microorganisms, toxins, and may be antibodies or lymphocytes traditionally, or even the most recent surface proteins and pathogen DNA, which are primarily used to prevent disease rather than to treat disease. Vaccine or vaccination is considered to be the most effective intervention for humans to control or eliminate many potentially fatal diseases and infections. Despite these achievements of vaccines, there is still space for improvement, such as:

  • Route of administration. Conventional needle injection is not so good in compliance.
  • There are no vaccines against numerous pathogens, particularly those intracellular pathogens such as human immunodeficiency virus and Hepatitis C virus.
  • Vaccines made from attenuated pathogens have safety concerns and risks, such as incomplete inactivation and possible mutation to pathogenic strains.

Lipid-Based Drug Delivery System Applied for Vaccines

To improve defects of current vaccines, pharmaceutical scientists are investigating adjuvants and novel vaccines delivery systems in recent years. The lipid-based delivery system is applied for vaccines with excellent advantages. And different delivery system is suitable for different vaccines, depending on physicochemical properties of antigens or antibody to be delivered.

  • Some lipid-based delivery systems are able to serve as both carriers and adjuvants for vaccines, such as virosomes and escheriosomes.
  • Lipid particulates can deliver antigens to antigen presenting cells more efficiently than soluble antigen to enhance cellular uptake.
  • Most lipid-based vesicles are flexible in structure and function, which can be tailored to adapt the system to various types of antigens, such as peptides, antigen-like proteins, and nucleic acids.
  • Antigens encapsulated into lipid-based vesicles or granules can protect the vaccine from being metabolized and destroyed by the digestive system, which makes oral administration possible.
