Pharmacosomes

Pharmacosomes, the colloidal dispersions of drugs covalently bound to lipids, may exist as ultrafine vesicular, micellar, or hexagonal aggregates depending upon the chemical structure of the drug-lipid complex. The criterion for the development of the vesicular pharmacosome is highly dependent on surface and bulk interactions of lipids with drug. Theoretically, any drug possessing an active hydrogen atom (- COOH, -OH, -NH2, etc.) can be esterified to the lipid, with or without spacer chain that strongly results in an amphiphilic compound.

Phaspholipids play major roles in drug delivery due to its amphiphilic nature that can modify the rate of drug release for the enhancement of drug absorption across biological barriers. The amphiphilic characters help pharmacosomes to reduce interfacial tension and at higher concentrations exhibit mesomorphic behaviour. Similar to other vesicular systems pharmacosomes provide an efficient method for delivery of both hydrophilic and lipophilic drugs directly to the site of infection, leading to reduction of drug toxicity with no adverse effects, also reduce the cost of therapy by improved bioavailability of medication especially in case of poorly soluble drugs.
