Introduction to NK Cell Membrane Coated Nanoparticles
NKM-NPs represent a sophisticated, bio-intelligent drug delivery platform. Their structure is a core-shell nanocarrier, where a synthetic inner core (polymeric, lipid, or inorganic NP) is cloaked by an extracted plasma membrane layer from active Natural Killer (NK) immune cells. The generation process involves three key stages: isolation of NK cell membranes via hypotonic lysis and homogenization, purification of the membrane fragments, and finally, self-assembly coating onto the pre-loaded nanoparticle core, often achieved through controlled high-pressure extrusion. This process effectively transfers the NK cell's complex surface proteins to the synthetic carrier.
