Background
AK4 is localized in the mitochondrial matrix and widely expressed in multiple types of tissues such as kidney, heart, and liver. It has been reported to be enzymatically inactive in vitro, but still retains its nucleotide-binding ability, physically interacts with the mitochondrial ADP/ATP translocator, and acts as a stress-responsive protein to maintain cell survival. The involvement of AK4 in the progression of multiple types of cancer has been investigated. AK4 has been associated with lung cancer and promoted the metastasis of lung cancer by downregulating ATF3.

According to previous studies, AK4 is involved in resistance to anti-tumor drugs via regulating mitochondrial activity and is a marker for metastasis and poor clinical outcome in lung cancer.
