Structural Organization of BCL10

BCL10 contains a CARD domain consisting of six tightly compressed alpha-helices and a C-terminal serine and threonine-rich functional domain. BCL10 protein covalently modifies phospholipid molecules on the plasma membrane through this domain, inducing phosphorylation, and recruiting card-related proteins to activate NF-κB.

CARD11-BCL10-MALT1 Complexes

CARD-BCL10-MALT1 (CBM) is a multiprotein signaling platform that controls most tissue immune and inflammatory pathways. These CBM regulatory mechanisms are essential for host defense and tissue homeostasis, and many genetic changes in CBM signaling components are associated with inherited and acquired immune-mediated diseases.

Inborn errors in each component of CBM are associated with a group of primary immunodeficiency disorders which include combined immunodeficiency, selective B-cell lymphocytosis, and atopic diseases, etc. Until now, most of our knowledge of the mechanism has come from CBM complexes in lymphocytes. Thus, further exploration of the physiological and pathological functions of CBM signaling is expected to provide additional insights into the environment-specific functions of immune and inflammatory pathways in health and disease.
