AKT subfamily is made up of three isoforms in mammals: AKT1, AKT2, and AKT3. AKT1 encodes the kinase B alpha (PKBα) protein which contains an N-terminal pleckstrin homology (PH) domain, a short α-helical linker, a kinase domain (KD), and a regulatory motif. MK-2206 is an orally available allosteric AKT1/2 inhibitor and BAY 1125976 is another AKT1/2 inhibitor that binds into an allosteric binding pocket formed by the kinase and PH domains of AKT1.

The E319G rs12881616, E17K rs121434592, L357P rs11555432, P388T rs11555431, rs2494732, and rs3803304 single nucleotide polymorphisms (SNPs) have been identified in the AKT1 gene.
