AKT2, known as RAC-beta serine/threonine-protein kinase, plays an important role in the conserved phosphoinositide 3-kinase signaling pathway, downstream of the insulin receptor. Like other AKT members, AKT2 is activated in cells in response to diverse stimuli such as hormones, growth factors, and extracellular matrix components. AKT2 mediates the physiological effects of insulin in several tissues including liver, skeletal muscle, and adipose tissue.

In humans, rare penetrant mutations in the AKT2 gene have been associated with monogenic disorders of glucose metabolism, such as p.Arg274His, p.Glu17Lys, and p.P50T. Alterations of AKT2 may contribute to the pathogenesis of ovarian carcinomas. Besides, AKT2 is also a useful molecular marker for predicting the prognosis of meningioma.
