Background

There are three mammalian isoforms of AKT: AKT1, AKT2, and AKT3 with AKT2 and AKT3 being approximately 82% identical to the AKT1 isoform. AKT3 is activated in cells exposed to stimuli such as hormones, growth factors, and extracellular matrix components. AKT3 phosphorylates and regulates the function of many cellular proteins involved in processes that include cellular metabolism, survival/apoptosis, and proliferation. AKT3 is the predominant AKT isoform in the adult mammalian brain and AKT3 gene mutations have been associated with a range of developmental disorders of the brain, including megalencephaly.

Analysis Services at Creative Biolabs

Creative Biolabs has raised a series of AKT3, phospho-AKT3 as well as anti-pan AKT antibodies which could be further used for custom AKT3 analysis assay development.

Besides traditional ELISA or WB, Creative Biolabs has already developed an improved assay for total (phosphorylated and unphosphorylated) AKT3 determination by utilizing a pair of matched antibodies that are highly specific for a distinct epitope on the AKT3 protein. In presence of AKT3 in a cell extract, the conjugates bring the donor fluorophore into proximity with the acceptor and thereby generate a FRET signal. Its intensity is directly proportional to the concentration of the AKT3 present in the sample. Moreover, sandwich ELISA system is available for the detection of AKT (Ser473).

On the other hand, studies demonstrated that knockdown of AKT3 can increase the metastatic potential of triple negative breast cancer cells. The results provide a rationale for the development of AKT isoform specific inhibitors.
