Background

T cell activation requires at least two signals to be fully activated. The first signal is provided by the interaction of T cell antigen-specific receptor and the antigen-major histocompatibility complex (MHC). The second signal is provided by co-stimulatory receptors like CD28. In engineered CAR-T cell, CAR consists of specific scFv and signaling endodomains of CD3ζ, CD28, and 4-1BB. Assay of Antigen Specific T Cell Activation

CAR-modified T cells can recognize tumor cells via binding of the CAR to its tumor-associated antigen (TAA) independent of TCR-MHC/peptide interactions, and the engineered signaling endodomains can help T cells be activated and persistent. As a result, T cells are activated and thus they can efficiently eliminate tumor cells by the secretion of perforin and granzymes as well as the expression of FasL and tumor necrosis factor-related apoptosis inducing ligand (TRIAL). In addition, other tumor-infiltrating immune cells can be activated by the secretion of various cytokines (see Fig. 1).
