Considerations of Choosing Successful Adjuvants

Adjuvant selection depends to a large extent on vaccine formulation and individual application. First, effectiveness and safety are critical in the assisted selection, but many other characteristics of successful adjuvants must be considered, including:

  • A stable, long shelf life compound;
  • Biodegradable;
  • Low production cost;
  • Does not induce an immune response against oneself;
  • Promote an appropriate immune response (i.e., cell or antibody immunity).

Adjuvants Type Description
Oil emulsions Usually considered too toxic to humans, oil emulsions such as Freund's complete adjuvant work by creating a depot at the injection site and slowly releasing antigens that trigger the desired immune response.
Bacterial-derived adjuvant Bacterial-derived molecules are a key source of immunostimulatory adjuvants, including lipopolysaccharide (LPS), which contains lipid A, an effective but toxic immunostimulant.
Carbohydrate adjuvant Dextran hydrochloride; mannan from Saccharomyces cerevisiae; chitosan from shrimp shell; β-D-glucan; dextran sulfate sodium salt derived from Leuconostoc.
Cytokine adjuvant Although the use of cytokine adjuvants is limited by toxicity and immunogenicity, they do have the potential to be DNA vaccines co-expressed with antigens. Many cytokines, such as IL-12, stimulate macrophage activation via TLR signaling.
Mineral salt adjuvants Mineral-derived compounds have been used for years as effective antigens, along with aluminum adjuvants. They are commonly referred to as Alum in literature and are widely used in human vaccines.
Other adjuvants Many vaccines use a combination of adjuvants to tailor a specific immune response. Squalene, decane, saponin, poly I:C and many other products have been found to be used alone or in combination with other adjuvants.
