PiggyBac Transposon System for CAR-T

Background

Viral vectors have been widely applied in traditional CAR-T cell therapy to treat genetic cancers. However, the maximum cargo size of viral vectors is restricted, thus hampering the transfer of larger designed CAR gene. Besides, safety and immunogenicity are also the major worries of the viral vectors. In addition, using living cells for the production of virus complicates the quality control and downstream processing. Fully understand new needs, Creative Biolabs has built up the non-virial PB transposon system to overcome these restrictions.

Introduction of PB Transposon System

There are two classes of transposons according to different mechanism of transposition, retrotransposons and DNA transposons. Among them, DNA transposons have become a research hotspot since they can transpose via a "cut-and-paste" mechanism, without creating a second copy of the DNA during transposition. PB transposon system belongs to DNA transposons. It is composed of a transposase (594 amino acids) and a transposon-like other transposon systems. The PB transposon is a movable genetic element that efficiently transposes between vectors and chromosomes. When performing transposition function, the PB transposase will recognize the transposon-specific inverted terminal repeats (ITRs) sequences which are located on both ends of the transposon vector. After that, the PB transposase moves the contents from its original positions and integrates them into TTAA chromosomal sites efficiently. In CAR gene delivery, the designed CAR genes are incorporated in the transposon vector. Of note, PB has the highest transposition activity in mammalian cells as well as the Sleeping Beauty (SB) transposon system.
