VivoCAR™ Anti-MICA (8C7) h(CD28-41BB-CD3z) CAR circRNA features the 8C7 clone, specifically optimized for its ability to block MICA shedding-a major mechanism of immune evasion. The importance of the 8C7 clone for in vivo CAR-T therapy lies in its dual action: direct cytolytic targeting and the preservation of membrane-bound MICA. The circular RNA architecture ensures a stable, high-level translation of the CAR, facilitating a potent therapeutic window that empowers the host's lymphocytes to overcome the soluble-ligand "smoke screen" used by metastatic tumors to disable immune surveillance.

  • Introduction
  • MICA is a stress-induced surface molecule expressed on epithelial and endothelial cells in response to cellular stress, infection, or malignant transformation. It is a ligand for the activating NKG2D receptor on NK cells and CD8+ T cells. Tumor cells often upregulate MICA but then shed it to evade immune surveillance. CAR-T cells targeting MICA can be used to directly kill MICA-expressing tumors or to target the soluble form of MICA to enhance NK cell activity.
