The biotin-binding immune receptors (BBIR) CAR use the biotin-avidin "lock-key" mechanism for CAR engineering. The BBIR containing dimeric avidin (dcAv BBIR) is used as the extracellular domain linked to an intracellular T cell signaling domain. Avidin was able to efficiently recognize and bind a variety of biotinylated antigen-specific molecules. The biotinylated antigen can be customized as scFv, mAb or tumor-specific ligand.
