Creative Biolabs provides an In Vivo CAR Safety Assessment Service designed to support safer and more predictable cell therapy development. This service addresses key translational challenges such as uncertain on-target/off-tumor toxicity, cytokine release risks, limitations of predictive animal models, and difficulties in evaluating biodistribution and long-term persistence of CAR-based therapeutics. By integrating preclinical in vivo modeling, multi-organ analysis, and quantitative pharmacology approaches, Creative Biolabs enables comprehensive safety evaluation of CAR-T and other engineered immune cell therapies.

Introduction

In vivo CAR safety assessment is a critical component in the development of CAR-T and CAR-NK therapies, ensuring therapeutic efficacy while minimizing systemic toxicity. Current research highlights key safety concerns such as cytokine release, neurotoxicity, and off-tumor effects, emphasizing the importance of comprehensive animal model evaluation. Integrated in vivo assessment strategies have become essential for supporting translational success and regulatory progression of engineered immune cell therapies.

What We Can Offer

Platform of One-stop In Vivo CAR Safety Assessment. (Creative Biolabs Original)

Our in vivo platform evaluates human CAR cell safety and efficacy through humanized mouse models that replicate true human immune dynamics. By utilizing custom transgenic models expressing human target antigens, we systematically screen for on-target/off-tumor toxicities across normal tissues.

Our Workflow

To launch the project, clients need to provide complete CAR construct design information covering target antigen and signaling domains, engineered immune cells or viral/non-viral delivery systems, alongside defined therapeutic indications and clear experimental research objectives for our analysis.

FAQs

Q: How to distinguish on-target and off-target toxicities on the platform?

A: It combines tissue-specific antigen profiling, human antigen-expressing transgenic models and multi-organ histopathology. CAR-induced damage to antigen-positive healthy tissues means on-target/off-tumor toxicity, while damage to antigen-negative tissues indicates off-target cross-reactivity.

Q: Can the workflow adapt to CAR-NK, CAR-Macrophage and other CAR therapies?

A: Yes. The optimized safety assessment protocols are compatible with various immune effector cells. All imaging, kinetic profiling and cytokine monitoring assays can be adapted to detect unique expansion, persistence and activation features of CAR-NK, CAR-M and other CAR therapies.

Q: What measures ensure the validity of CRS modeling?

A: Standardized humanized mouse cohorts with verified human cell reconstitution are used to reduce model variability. Intensive blood sampling is conducted post-cell injection to capture transient cytokine spikes and ensure accurate CRS modeling.
