Conventional CAR-T therapies face critical hurdles including persistent genomic integration risks, lengthy ex vivo manufacturing timelines, and suboptimal in vivo targeting efficiency. To address these limitations, we have established our mRNA-LNP Platform, which leverage non-integrating mRNA technology combined with advanced nanoparticle encapsulation. Our service provides complete solutions encompassing mRNA design, synthesis, formulation, and functional characterization, delivering fully assembled, targetable nanocarriers tailored to specific T cell subsets. By enabling rapid, flexible, and off-the-shelf compatible production with precise antibody directed delivery, we empower researchers and clinicians to accelerate preclinical development while significantly reducing safety concerns and manufacturing complexity.
Messenger RNA encapsulated in lipid nanoparticles (mRNA-LNPs) represents a transformative non-viral platform for transient protein expression, combining high translational efficiency with a favorable safety profile due to its cytoplasmic activity and lack of genomic integration. In CAR therapy, mRNA-LNPs offer a paradigm shift by enabling ex vivo or even in vivo delivery of CAR-encoding transcripts to immune effector cells, thereby circumventing the protracted manufacturing, high costs, and insertional mutagenesis risks associated with conventional viral vectors.

Our platform supports swift and economical engineering of primary or endogenous immune cells, bypassing viral integration and permanent genomic alterations. Through the synergistic use of proprietary ligand targeted LNPs, endosomal release promoters, and adaptable mRNA cassettes, we achieve potent and reversible receptor expression with a favorable safety profile. This system facilitates high throughput preclinical evaluation, enables bespoke therapeutic design, and accommodates scalable production workflows.
Our platform integrates targeted lipid nanoparticle delivery, endosomal escape engineering, customized mRNA design, and optimized electroporation protocols to enable rapid, flexible, and scalable transient CAR expression in both endogenous and primary immune cells for next generation immunotherapy development.
We prioritize mRNA-LNP based CAR production for its non-integrating safety profile, rapid and flexible manufacturing timeline, and precise tunability of transgene expression. Our platform supports iterative receptor screening, consistent quality control, and versatile application in both autologous and allogeneic settings, collectively positioning it as a powerful and clinically adaptable tool for next-generation T cell immunotherapy.
mRNA technology precludes genomic integration risk, ensuring high safety. It supports direct in vivo production, improving accessibility, while its rapid synthesis and antibody mediated targeting enable flexible, precise delivery to specific T cell subsets.
Q1: What is the fundamental difference between mRNA-LNP and viral vector-based CAR/TCR engineering?
A1: mRNA-based engineering is non-integrating. It delivers genetic instructions into the cytoplasm for transient protein expression without altering the host genome, whereas viral vectors permanently integrate the CAR/TCR gene into the T cell's DNA, carrying a risk of insertional mutagenesis.
Q2: How long does the CAR/TCR transgene expression last, and is it sufficient for therapeutic effect?
A2: Expression is transient, typically peaking within 24-48 hours and lasting for several days. For many applications, especially where a "controlled pulse" of activity is needed, this transient window is sufficient and advantageous for safety. For malignancies requiring prolonged effector function, repeated dosing is being explored.
We deliver next-generation CAR/TCR platforms leveraging transient mRNA expression to ensure unparalleled safety, eliminating genotoxic risk and enabling controlled, reversible immune modulation. Our advanced encapsulation technologies, including targeted LNPs, preserve T cell fitness while facilitating scalable, off-the-shelf accessibility, positioning your programs at the forefront of precision immunoengineering.
"Adopting Creative Biolabs' mRNA-LNP transient CAR expression and encapsulation system has fundamentally reshaped our risk benefit assessment. The tightly regulated expression window provides a pharmacologically reversible off switch for receptor activity following infusion, an especially valuable feature in autoimmune indications, where short-term modulation is clinically favoured over permanent ablation." Dr. A. R.*
"This platform enables us to separate CAR expression from viral transduction, effectively removing both the scheduling uncertainty and the regulatory complexities associated with lentiviral vector manufacturing. Across three independent production runs, we achieved consistently high encapsulation rates, with minimal batch-to-batch variation in particle size and RNA integrity." Dr. M. T.*
