Despite the clinical success of CAR-T therapies, durable antitumor activity remains constrained by transient transgene expression, genotoxic risks associated with integrating vectors, and the high manufacturing costs of ex vivo cell engineering. Our saRNA-LNP platform addresses these limitations by enabling sustained, non-integrative CAR expression through a fully synthetic, repeat-dose–compatible delivery system. We offer a comprehensive service from saRNA construct design and LNP formulation to in vivo functional validation. Our solution ensures prolonged CAR surface presentation, reduced immunogenicity, and flexible redosing, empowering you with off-the-shelf, cost-effective tools for next-generation cell immunotherapy development.
Self-amplifying RNA (saRNA) encapsulated in lipid nanoparticles (LNPs) represents a next-generation nucleic acid delivery platform that enables prolonged, high-level transgene expression with substantially reduced dosing frequency compared to conventional mRNA. saRNA-LNP provides a compelling alternative to viral vectors and transient mRNA electroporation by sustaining chimeric antigen receptor expression over extended periods, thereby enhancing cytotoxic persistence, reducing T cell exhaustion, and potentially improving long-term therapeutic outcomes in adoptive cell transfer regimens.

Our proprietary saRNA-LNP Platform for Sustained CAR Expression combines self-amplifying RNA with advanced lipid nanoparticles, enabling robust, multi-week CAR expression from a single, ultra-low dose without genomic integration risks. We provide biopharma partners with customizable, high-efficiency formulation and targeting solutions, significantly accelerating the development of safer, more cost-effective, and highly scalable ex vivo and in vivo CAR therapeutics.
Our platform offers integrated saRNA vector engineering from backbone selection and base modification to UTR/poly(A) optimization paired with comprehensive functional validation, enabling sustained, potent CAR expression and long-lasting anti-tumor activity.
Q1: How does the saRNA-LNP platform achieve sustained CAR expression compared to conventional mRNA?
A1: Conventional mRNA degrades rapidly, leading to transient CAR expression. Our saRNA platform incorporates engineered viral non-structural protein genes. Once delivered into the cytoplasm via LNPs, these proteins form a replicase complex that continuously amplifies the CAR-encoding RNA template, extending CAR expression from days to several weeks.
Q2: Are there any genomic integration risks associated with the saRNA platform?
A2: No. The entire self-amplification and translation process occurs strictly within the host cell cytoplasm. Unlike lentiviral or retroviral vectors, saRNA does not enter the nucleus and does not integrate into the host genome, eliminating the risk of insertional mutagenesis.
Our cutting-edge saRNA-LNP platform redefines CAR cell therapy by delivering sustained, multi-week receptor expression alongside profound dose-sparing efficiency via autonomous cytoplasmic self-replication. By enabling precise multigenic co-expression and advanced logic gating without genomic integration risks, we provide an ultra-safe, highly scalable, and cost-effective solution tailored for next-generation oncology therapeutics.
"We were looking for a way to achieve the prolonged therapeutic window of viral CAR-T therapies without the associated insertional mutagenesis risks. This saRNA-LNP platform exceeded our expectations. The self-amplifying RNA sustained robust CAR expression in primary T cells for over two weeks from a single transfection, significantly outperforming standard mRNA." Dr. Alistair Vance.
"The synergy between their high-yield saRNA synthesis and optimized LNP encapsulation is outstanding. We utilized this service for in vivo CAR-T generation and observed exceptional spleen and lymph node targeting with minimal systemic toxicity. The LNP stability and uniform particle size meant we got highly reproducible CAR expression dynamics across all test cohorts." Dr. Elena Rostova.
