Creative Biolabs provides an advanced AdCAR-T Development Service for Antigen Re-Targeting, designed to address key challenges such as antigen escape, tumor heterogeneity, and on-target/off-tumor toxicity. By utilizing an innovative modular split-component receptor design, our platform enables precise and flexible multi-antigen targeting, significantly improving safety and therapeutic efficacy. This next-generation solution empowers researchers to overcome antigen–loss–driven relapse and accelerate the development of adaptable CAR-T therapies for solid tumors.
Adapter CAR-T (AdCAR-T) technology addresses the fundamental limitations of single-specificity CARs by employing a split recognition/activation design. This system engineers T cells to recognize a universal label rather than a tumor antigen. A separate, non-engineered adapter molecule (AM) acts as the bridge, binding the tumor antigen and the AdCAR-T cell simultaneously.

The core value of our AdCAR-T system is its flexibility and control. Unlike conventional, fixed-target CARs, our platform separates T-cell activation from antigen recognition. This allows a single, universally engineered T-cell product to be redirected against any target simply by changing the external AM.
We provide a direct solution to the two biggest challenges in cell therapy:

AdCAR-T cells employ a universal CAR structure that binds to a customizable adaptor molecule. This adaptor molecule bridges the CAR-T cell and the tumor cell, carrying both an antigen-specific antibody fragment and a tag recognized by the AdCAR.

To initiate an AdCAR-T development project, clients typically need to provide target antigen information for 2-3 Tumor-Associated Antigens (TAAs), the sequence for the adapter molecule, and a source of T-cells for engineering and validation.
The final project deliverables consist of three primary components: a comprehensive AdCAR vector report, a validated AdCAR-T cell line with a complete data package, and a detailed adpter molecule specification report.
How does AdCAR-T compare to bispecific or tandem CARs?
Unlike fixed bispecific or tandem CARs, AdCAR-T is dynamically re-targetable by switching the AM, enabling immediate adaptation to antigen escape without re-engineering the T cells.
What is the key safety advantage of AdCAR-T?
Activity is controlled by AM dosage. Stopping AM infusion acts as a rapid "off-switch", offering reversible control to manage severe toxicities.
Can I use my proprietary antibodies as AMs?
Yes. We conjugate and characterize your proprietary antibodies or fragments into validated Adapter Molecules, integrating them seamlessly into our platform.
What is the expected persistence of AdCAR-T cells?
AdCAR-T cells are built on persistent backbones and remain resting without constant stimulation, supporting long-term readiness and reduced exhaustion.
