While conventional CAR-T therapies offer transformative potential, their development remains severely constrained by the high production costs, limited payload capacity, and complex safety profiles of viral vectors. To overcome these hurdles, we offer our advanced SB Transposon Platform for Non-Viral CAR-T Development. Our comprehensive service spans from custom transposon vector design and optimization to highly efficient in vitro T cell engineering and rigorous functional validation. By leveraging our scalable and cost-effective system, we empower you to rapidly accelerate pre-clinical pipelines, enhance therapeutic safety, and successfully advance novel immunotherapies toward clinical application.
The Sleeping Beauty (SB) transposon system comprises a synthetic DNA transposase capable of precisely integrating therapeutic transgenes into vertebrate genomes, thereby overcoming the limitations of persistent episomal expression. SB mediates cut-and-paste transposition via a bipartite recognition mechanism, wherein the transposase binds to inverted terminal repeats (ITRs) flanking the gene of interest, excises the cassette from a donor plasmid, and integrates it into TA-dinucleotide-rich chromosomal loci with a defined, yet relatively random, genomic distribution.

Our SB Transposon Platform offers a non-viral solution for CAR-T development, overcoming the cost, safety, and payload constraints of traditional viral systems. By enabling stable transgene integration and sustained CAR expression, it streamlines T cell engineering from design through validation. We deliver scalable, cost-effective manufacturing and accelerated preclinical timelines, empowering you to advance safer immunotherapies to the clinic with confidence.
This high-performance transposition system, belonging to the Tc1/mariner superfamily. Optimizing non-viral CAR-T development, it combines an exceptional cargo capacity with highly efficient human cell integration and a safer, nearly random insertion profile.
Q1: How does the genomic integration profile of the SB transposon differ from lentiviral vectors, and what are the safety implications?
A1: Unlike lentiviruses that preferentially target transcriptionally active regions and risk oncogene activation, the SB system utilizes a "cut-and-paste" mechanism targeting TA dinucleotides near-randomly. Avoiding gene-dense areas significantly minimizes insertional mutagenesis and T cell malignant transformation, delivering a superior clinical safety profile for cell therapy development.
Q2: What is the cargo capacity limit of the SB transposon platform, and does it support multi-targeted CAR engineering?
A2: While lentiviral vectors are constrained to under 9 kb, the SB platform efficiently accommodates inserts exceeding 10-11 kb without compromising integration efficiency. This expanded capacity easily supports complex, multi-targeted CAR designs and the co-expression of immunomodulators like IL-15 or PD-1 checkpoints to remodel the solid tumor microenvironment.
Our SB Transposon Platform delivers a highly efficient, cost-effective, and scalable non-viral alternative for CAR-T cell manufacturing. By ensuring high-fidelity gene integration, robust therapeutic expression, and minimized immunogenicity, we streamline the path from discovery to clinical translation.
"Switching to this SB Transposon Platform revolutionized our pipeline. We completely bypassed the lengthy and expensive viral vector manufacturing bottlenecks. The platform delivered exceptionally high integration efficiency and stable CAR expression, allowing us to accelerate our preclinical timelines by months." Dr. A* K*.
"Safety and scalability were our primary concerns. This platform's non-viral approach significantly reduced the risk of insertional mutagenesis while maintaining a highly favorable safety profile. The cell viability post-transfection was outstanding, ensuring robust expansion for our downstream applications." Prof. E* D*.
