Background Platforms What We Can Offer Workflow Highlights FAQs Contact

Creative Biolabs provides a Helper-Dependent Adenoviral (HDAd) Platform for CAR Delivery to support the development of next-generation cell and gene therapies. This platform addresses key challenges including limited cargo capacity of AAV vectors, integration-associated genomic risks of lentiviral systems, and immune clearance concerns associated with conventional adenoviral vectors. Through high-capacity gutless adenoviral vector engineering and optimized packaging strategies, Creative Biolabs enables delivery of complex, multi-target CAR designs and supports non-integrating CAR expression approaches.

Introduction

Helper-Dependent Adenoviral (HDAd) vectors, also termed "gutless" or high-capacity adenoviruses, contain only viral inverted terminal repeats (ITRs) and a packaging signal. By removing all viral genes, HDAd vectors eliminate acute immune clearing seen in early-generation adenoviruses while maintaining superior transduction efficiency across dividing and post-mitotic cells. With an expansive cloning capacity (~37 kb), HDAd vectors effortlessly accommodate multi-gene circuits, synthetic switches, and multi-armored CAR systems. Advanced tropism modifications, such as HSC-targeting fiber variants, position HDAd as a premier non-integrating platform for next-generation in vivo CAR cell engineering.

Our Platforms

Creative Biolabs delivers customized HDAd platforms specifically tailored for complex synthetic biology and CAR engineering projects. By eliminating all viral protein-coding genes, our gutless adenoviral vectors offer an unprecedented payload capacity of up to ~37 kb, allowing researchers to incorporate multi-antigen targeting CARs, inducible safety switches, multiplexed cytokine armoring modules, and sophisticated logic-gate circuits within a single vector construct. Our key capabilities and client deliverables include:

  • High-Capacity Vector Construction: Seamless assembly of complex transgenes, regulatory elements, and dual/multi-CAR expression cassettes without payload size limitations.
  • Non-Integrating Genetic Modification: Safe transient to long-term episomal transgene expression in non-dividing and dividing immune cell populations, avoiding insertional mutagenesis risks.
  • Low Immunogenicity Engineering: Complete removal of viral coding regions minimizes host cytotoxic T lymphocyte (CTL) responses, supporting sustained therapeutic activity.
  • Tailored Cell Tropism: Pseudotyping options (such as fiber modification for targeting hematopoietic stem cells and immune cell lineages) for direct in vivo CAR-T and CAR-NK vector applications.

FAQs

Q1: Why choose HDAd over Lentivirus or AAV for CAR delivery?

A1: While lentiviruses integrate randomly into host genomes and AAVs are restricted to small cargos (~4.7 kb), HDAd offers a massive ~37 kb payload capacity and non-integrating episomal expression. This permits delivery of multi-armored CARs, logic gates, and safety switches in a single, non-mutagenic vector.

Q2: How does Creative Biolabs manage helper virus contamination in HDAd lots?

A2: We utilize optimized Cre/loxP recombination packaging systems coupled with advanced chromatography and gradient ultracentrifugation steps. Our analytical testing suite includes highly sensitive quantitative PCR assays to ensure helper virus levels remain below rigorous research thresholds.

Q3: Can HDAd vectors be used for direct in vivo CAR-T or CAR-NK applications?

A3: Yes. By utilizing chimeric fiber variants, our platform can selectively target surface markers on hematopoietic stem cells and lymphoid lineages, facilitating in vivo primary immune cell reprogramming without requiring ex vivo cell handling.
