The anti-WT-1 chimeric antigen receptor (CAR) THP1 cell line is a stable cell line developed using anti-WT-1 CAR lentivirus transfection. This transfected CAR lentiviral vector is constructed to express a scfv of anti-WT-1 antibody (6F-H2) linked to CD3ζ signaling domains.The anti-WT-1 CAR engineered THP1 cells can be used to be differentiated into macrophage cells for in vitro or in vivo phagocytosis studies. This CAR-THP1 cell line product can be used for the researches of Ovarian cancer.

  • THP-1 is a human leukemia monocytic cell line, which has been extensively used to study monocyte/macrophage functions, mechanisms, signaling pathways, and nutrient and drug transport. This cell line has become a common model to estimate modulation of monocyte and macrophage activities. CAR engineered THP1 cells can be differentiated into macrophages using PMA (Phorbol 12-myristate 13-acetate). Once differentiated (M0 macrophages), they were incubated with IL-4 and IL-13 in order to obtain M2 polarized macrophages or with IFN-gamma and LPS for classical macrophage activation (M1).

  • CD3ζ:
    CD3ζ is used as the CAR intracellular signaling domain. CD3ζ has homology to the Fc common γ-chain, FcεRI-γ, a canonical signaling molecule for antibody_x005f_x005f_x005F_x005f_x005F_x0002_dependent cellular phagocytosis (ADCP) in macrophages. CD3ζ, also known as T-cell receptor zeta, which together with T-cell receptor and CD3γ, δ , ε chain, forms the TCR-CD3 complex. ζ was expressed independently from the complex. The zeta chain plays an important role in coupling antigen recognition to several intracellular signal-transduction pathways. CD3-zeta, which contains 3 ITAMs, is the most commonly used endodomain component of CARs. It transmits an activation signal to the T cell after antigen is bound. CD3-zeta may not provide a fully competent activation signal and additional co-stimulatory signaling is needed. For example, chimeric CD28 and OX40 can be used with CD3-zeta to transmit a proliferative/survival signal, or all three can be used together.

  • Introduction
  • This gene encodes a transcription factor that contains four zinc-finger motifs at the C-terminus and a proline/glutamine-rich DNA-binding domain at the N-terminus. It has an essential role in the normal development of the urogenital system, and it is mutated in a small subset of patients with Wilms tumor. This gene exhibits complex tissue-specific and polymorphic imprinting pattern, with biallelic, and monoallelic expression from the maternal and paternal alleles in different tissues. Multiple transcript variants have been described. In several variants, there is evidence for the use of a non-AUG (CUG) translation initiation codon upstream of, and in-frame with the first AUG. Authors of PMID:7926762 also provide evidence that WT1 mRNA undergoes RNA editing in human and rat, and that this process is tissue-restricted and developmentally regulated.
