CAR T-cell Screening

Screening CAR-T cells is important for developing safe, effective, and personalized therapies. It allows for the evaluation of potential side effects or adverse reactions of CAR-T cell therapy. Screening helps determine the effectiveness of CAR-T cells in targeting and killing target cells. It also helps tailor CAR-T cell therapy, considering factors such as tumor type, genetic profile, and immune response. The screening process of CAR-T cells requires strict control to ensure the accuracy and effectiveness of CAR design and transfection.

Organ-On-A-Chip Platform for CAR T-cell Screening

Organ-on-a-chip is a small functional unit able to simulate the level of the organ in vitro. CellRapeutics™ organ-on-a-chip platform allows researchers to observe how the CAR-T cells interact with the surrounding cells and tissues, assess their cytotoxicity, and evaluate their efficacy in a controlled microenvironment.

Organ-on-a-chip devices allow researchers to control key parameters such as cell types, cell-cell interactions, and chemical gradients, enabling precise manipulation of the T-cell environment. Additionally, researchers can reduce the reliance on animal models for T-cell screening by using the organ-on-a-chip platform.

Screening CAR-T cells through the organ-on-a-chip platform typically involves the following points:

  • Establishment of Organ-On-A-Chip Model: Develop an organ-on-a-chip model that mimics the microenvironment of the target tissue or organ where the CAR-T cells will be tested.
  • Integration of CAR-T Cells: Introduce CAR-T cells into the organ-on-a-chip model either by direct seeding or perfusion to allow interaction with the organotypic environment.
  • Functional Assessment: Monitor the behavior and function of CAR-T cells within the organ-on-a-chip model, including their interaction with target cells or tissues and their cytotoxic activity.
  • Drug Response: Evaluate the response of CAR-T cells to specific drugs or treatments within the organ-on-a-chip platform to assess their efficacy and potential side effects.
  • Real-time Monitoring: Utilize imaging techniques or sensors within the organ-on-a-chip system to track the dynamics of CAR-T cell behavior and response in real time.
  • Data Analysis: Analyze the data collected from the organ-on-a-chip experiments to assess the performance and suitability of CAR-T cells for targeted therapy.
