VivoCAR™ Anti-CA9 (M75) h(CD28-41BB-CD3z) CAR circRNA features the M75 clone, recognized for its high-affinity binding to the proteoglycan-like domain of CA9. For in vivo applications, this clone is essential for its ability to distinguish malignant cells within hypoxic tumor zones. The circRNA architecture protects the transcript from degradation, facilitating sustained CAR expression on endogenous T cells. This allows for a robust, localized attack on the pH-regulating machinery of solid tumors, effectively neutralizing a key survival mechanism of aggressive carcinomas in a non-integrating format.

  • CA9 is a hypoxia-inducible, membrane-bound enzyme that is highly expressed in the hypoxic regions of many solid tumors (such as Renal Cell Carcinoma). CA9 helps maintain an acidic tumor microenvironment, promoting tumor growth and metastasis. Its restricted expression in normal tissues makes it an excellent target for CAR-T cell therapy.
