Introduction

T Cell Antigen Coupler (TAC) T cells represent a promising alternative to conventional CAR-T therapies, as they leverage the complete endogenous T cell receptor (TCR) signaling complex to drive tumor cell killing while minimizing cytokine release and avoiding tonic signaling. The clinical application of cell therapies for solid tumors, however, is hampered by insufficient T cell infiltration, poor persistence, and a suppressive tumor microenvironment. A novel NECTIN-4-redirected TAC-T cell variant, NECTIN-4 TAC28-T, engineered with the intracellular co-stimulatory CD28 domain to enhance its anti-tumor potency.
