CAR Expression Detection for CAR-MA Therapy

The success of CAR expression is an important step in the development of CAR-based cell therapy. We provide accurate and reproducible in vitro and in vivo CAR expression detection methods.

Background

CARs are genetically engineered receptors that can recognize specific antigens and subsequently activate downstream signaling. These receptors can be artificially expressed in human T cells, NK cells, or macrophages using gene transfer techniques, such as lentiviral transduction. CAR-MA kills tumor cells through CAR targeting and exerts targeted phagocytosis, as well as remodeling the tumor microenvironment by inducing inflammation and recruiting T cells.

Evaluating CAR expression is an essential step in the production of CAR cells. This is often done by flow cytometry, using protein L, anti-Fab antibodies or target antigens as detection methods. Among these common choices, target antigens are widely considered to be the best option, because it offers high specificity and minimal background staining. In vitro visualization methods, including confocal and total internal reflection fluorescence microscopy, have captured the molecular details underlying CAR immunological synapse formation, signaling, and cytotoxicity. An important step for CAR-MA therapy is to evaluate CAR expression in vivo. In vivo tracking methods, including two-photon microscopy, bioluminescence imaging (BLI), and positron emission tomography scanning (PET), have been used to monitor CAR-cells' biodistribution across blood, tissue, and tumor.
