What is avidity?

The binding events between T cells and their target tumor cells determine the start of immune synapse formation and T cell activation. Therefore, cell-cell interaction is a key parameter to consider when trying to understand the T cell response process. Cell avidity defines the total intercellular force between multiple parallel interactions, including co-receptor binding, TCR clustering, cell adhesion proteins, even orientations, and valences.

Compared with affinity, cell avidity provides a more complete and physiologically relevant image, reflecting the true interaction between effector cells and tumor cells. These interactions help to better predict cellular responses and outcomes during immunotherapy.

Why cell avidity?

The field of cell therapy lacks a proper understanding of well-defined therapies. Currently, the methods commonly used to select the best cell therapy effector cells include surface plasmon resonance (SPR) affinity studies, tetramer staining, and functional determination. However, the results of affinity and tetramer determination are inconsistent, and there is no linear relationship with immune cell response. Functional assays such as IFN-γ secretion and cell killing are more predictive of in vivo effects, but they are time-consuming and inconsistent between experiments or assays.

Through cell avidity measurement, researchers can study cellular interaction characteristics corresponding to immune cell responses in a predictable, reproducible, and rapid manner. All these analyses are performed at a high-throughput and single-cell level without compromising cell viability.

How does T-AviA™ work?

The functional avidity is determined by exposing the T cell population to different amounts of homologous antigens in vitro. T cells with high functional affinity respond to low antigen doses. It is believed that this in vitro measurement correlates well with the in vivo effector capacity of T cells. The core of T-AviA™ technology is the piezoelectric element that generates resonant sound waves. These ultrasounds are the basis of cell affinity analysis because they can pull the cell vertically to the first acoustic node where the force is zero. This seamless method of separating interacting cells allows you to directly measure and record the cell-specific affinity between effector cell populations.

We optimized the workflow of T-AviA™ to promote the user experience and provide you with reliable, repeatable, and fast results. The T-AviA™ chip is specifically designed to simplify cell culture and keep samples under physiological conditions when performing experiments. Once an analysis has been performed, a new batch of effector cells can be simply flushed-up to 5 times!
