γδ T cells are a small subset of peripheral blood T lymphocytes and can function as a promising tool for antitumor immunotherapy. Creative Biolabs is dedicated to addressing the challenges of isolating and expanding γδ T cells and enabling the parallel development of multiple allogeneic cell therapies for hematological malignancies and solid tumors.

Introduction

γδ T cells are unique unconventional T cells and are tissue-resident immune cells with inherent anti-tumor functions that do not rely on the expression of tumor-specific antigens and therefore provide a promising target for CAR cell therapy. However, these cells represent a small fraction (1-5%) of the peripheral T-cell pool and require activation and propagation to achieve clinical benefit. Typically, elaborate purification protocols such as FACS-based cell sorting or expansion in culture are needed to obtain enough γδ T cells for subsequent studies. Now we provide an available approach to isolate and purify γδ T cells from peripheral blood mononuclear cells (PBMC).

Isolation of γδ T cells

Purification of γδ T cells is needed for insight when studying their anti-cancer functionality and their utilization in adoptive cell therapy. Recent clinical trials have included approaches where magnetic labeling systems and column isolation were used for the selection of purified TCR γδ-expressing T cells. We can offer services to isolate the γδ T cells from PBMC by magnetic antibody cell sorting (MACS) technology. γδ T cells can be purified by positive or negative magnetic selection using the micro-Bead. We also provide γδ T cells isolation kit. By using isolation kits, the purity of these cells is more than 90% after separation from PBMCs. The γδ T cells are rested overnight in the medium before using them for further experiments.

  • TCRγδ+ T cells are indirectly magnetically labeled with anti-TCRγδ micro-bead. The magnetically labeled TCRγδ+ T cells are retained on the column and eluted after removal of the column from the magnetic field.
  • The non-γδ T cells are indirectly magnetically labeled with a cocktail of biotinylated monoclonal antibodies and anti-biotin microbeads. Magnetic labeling of nonTCRγδ+ T cells so that γδ T cells are isolated by “negative selection”. The magnetically labeled, unwanted cells are subsequently depleted by separation over a MACS column while the unlabeled γδ T cells pass through the column.
