Summary: This study explored a novel nanoparticle platform for efficiently expanding antigen-specific CD4+ T cells in vitro. By mimicking the function of antigen-presenting cells (APCs), these nanoparticles, displaying major histocompatibility class II (MHC II) molecules, effectively stimulated the growth of CD4+ T cells, including rare subsets, when combined with soluble costimulatory signals. This approach demonstrated the potential to overcome limitations in current CD4+ T cell expansion techniques, paving the way for more consistent and scalable CD4+ T cell-based therapies and facilitating deeper investigations into CD4+ T cell biology.
