Cytochrome c-specific TCR 226 has a very different sensitivity to the I-Ek mutants when it recognized the wild-type MCC-p5K versus MCC-p5E. These different patterns of sensitivity suggested that 226 was binding in very different orientations depending on the ligand. 226 has a much higher affinity,> 1000-fold, for MCC/I-Ek than for the p5E variant.
