  • Annexins are a series of calcium ion-dependent phospholipid binding proteins that preferentially bind phosphatidylserine (PS). Under normal physiological conditions, PS is mainly located on the inner layer of the plasma membrane. After the start of apoptosis, the PS gradually disappeared through the phospholipid bilayer, and transferred to the outer layer of the cell membrane, marking the cell as a target for phagocytosis. On the outer surface of the cell membrane, the PS can be detected in the calcium ion-dependent mode by fluorescently labeled Annexin V.

    In the early stage of apoptosis, the plasma membrane can block reactive dyes such as propidium iodide (PI), 7-AAD, or Fixable Viability Dye such as eFluor™ 660 or eFluor™ 780 from entering the cell. These cells will be stained by Annexin V, but not by reactive dyes, which can distinguish cells in the early stage of apoptosis. However, in the late stage of apoptosis, the cell membrane loses its integrity, so Annexin V can also contact the intracellular PS. Reactive dyes can be used to distinguish these late apoptosis and necrotic cells (Annexin V, reactive dye positive) from early apoptotic cells (Annexin V positive, reactive dye negative).
