VivoCAR™ Anti-HMW-MAA (763.74) h(CD28-41BB-CD3z) CAR circRNA utilizes the 763.74 clone to target the chondroitin sulfate proteoglycan-4 (CSPG4) on melanoma cells and the tumor vasculature. This clone is significant for its ability to simultaneously destroy tumor cells and disrupt the angiogenic support system. By delivering this CAR via the circRNA platform, we enable a potent anti-tumor surge. The non-integrating nature of the transcript allows for precise control over the treatment duration, providing a safe method to target the melanoma-associated stroma without permanent genetic modification.

  • HMW-MAA is a large membrane-bound chondroitin sulfate proteoglycan. It is highly and universally expressed on the surface of most Melanoma cells (over 90%), as well as cells from neuroectodermal origin, but is largely restricted in expression on normal adult tissues. Its consistent and high expression on the cell surface makes it a historically and continuously relevant target for immunotherapy against melanoma, which is typically a difficult-to-treat solid tumor.
