The vector of anti-mesothelin chimeric antigen receptor (CAR) is constructed for the engineering of T cells to target human mesothelin. The T cells are genetically modified through transduction with a lentiviral vector expressing scFv of anti-mesothelin antibody linked to KIR2DS2, and another vector expresssing DAP12 signaling domain. When introduced into human T cells with DAP12, the KIR-based CAR triggers enhanced antigen-specific proliferation and anti-tumor activity. And the vector product was designed for the treatment of pancreatic cancer, ovarian cancer, lung cancer, malignant pleural mesothelioma tumors (MPM tumors).

  • KIR2DS2
    Killer cell immunoglobulin-like receptors (KIRs) are transmembrane glycoproteins expressed by natural killer cells and subsets of T cells. The KIR proteins are classified by the number of extracellular immunoglobulin domains (2D or 3D) and by whether they have a long (L) or short (S) cytoplasmic domain. KIR proteins with the short cytoplasmic domain lack the ITIM motif and instead associate with the TYRO protein tyrosine kinase binding protein to transduce activating signals.KIR proteins are thought to play an important role in regulation of the immune response.
    DAP12
    This protein may associate with the killer-cell inhibitory receptor (KIR) family of membrane glycoproteins and may act as an activating signal transduction element. It may bind zeta-chain (TCR) associated protein kinase 70kDa (ZAP-70) and spleen tyrosine kinase (SYK) and play a role in signal transduction, bone modeling, brain myelination, and inflammation.
