VivoCAR™ Anti-GPRC5D (2A4) h(CD28-41BB-CD3z) CAR circRNA utilizes the 2A4 clone to target GPRC5D, a lineage-specific surface receptor highly prevalent in multiple myeloma. The significance of the 2A4 clone for in vivo applications lies in its capacity to identify malignant plasma cells with exceptional specificity, effectively bypassing common B-cell antigens. By utilizing the circRNA platform, this product generates a transient surge of CAR activity that concentrates the immune response within the bone marrow niche. This approach minimizes the risk of prolonged cytopenia and allows for the controlled, non-permanent eradication of myeloma cells.

  • GPRC5D is a highly restricted G-protein-coupled receptor characterized by seven transmembrane domains. It exhibits dense and specific expression on the surface of malignant plasma cells in Multiple Myeloma (MM), while its presence in healthy tissues is strictly localized to hard keratinized structures, such as hair follicles. This distinct expression pattern establishes it as an ideal "next-generation" target for MM, particularly for patients who have relapsed following BCMA-targeted therapies, given that GPRC5D and BCMA are expressed independently of one another.
