VivoCAR™ Anti-MUC16 (11D10) h(CD28-41BB-CD3z) CAR circRNA focuses on targeting the proximal extracellular domain of MUC16, which remains on the cell surface after the CA125 fragment is shed. For in vivo applications, the 11D10 clone is critical for circumventing the "decoy effect" of circulating soluble antigens. The circRNA platform ensures a stable and durable expression of the CAR on endogenous lymphocytes, facilitating the persistent destruction of residual ovarian cancer cells in the peritoneal cavity without the risks associated with permanent viral gene delivery.

  • MUC16 is a very large, heavily glycosylated mucin protein highly expressed on ovarian cancer cells and several other adenocarcinomas. The shedding of its extracellular domain is measured as CA-125 in serum. The large, extracellular domain provides numerous accessible epitopes, making it a well-established and essential target for CAR-T cell therapy, especially for peritoneal malignancies.
