MACF1, a protein scaffold of more than 700 kDa, coordinates the crosstalk between actin microfilaments and microtubules to support cell-cell connections, cell polarity, vesicular transport, proliferation, and cell migration. Structurally, MACF1 comprises a variable N-terminal domain, central region composed of a plakin domain and a spectrin repeat-containing rod domain and a C-terminal MT-binding domain.
Six different MACF1 isoforms produced via alternative splicing and differential promoter usage have been identified, including MACF1a1, MACF1a2, MACF1a3, MACF1-4, MACF1b, and MACF1c. Unique arrangements of these motifs by RNA splicing or transcription start site variation confers to each isoform its own functional characteristics.

Aberrant expression of MACF1 is also related to various human cancers. Further investigation using proteomics techniques identified MACF1 as a potential oncofetal biomarker that may be used for the diagnosis of colorectal cancer, pancreatic cancer, non-small cell lung cancer.
