ABCG5 is a transport protein that functions as a half-transporter to limit intestinal absorption and promote the biliary excretion of sterols. ABCG5 and ABCG8 genes encode for two proteins sterolin-1 and -2, respectively. Mutations in either genes result in a lipid disorder, sitosterolemia.
Sitosterolemia is a rare inherited disorder characterized by increased levels of plasma plant sterols (PS) such as stigmasterol, campesterol, and sitosterol. A high level of plasma PS would lead to premature CVD, accelerated atherosclerosis, and sudden cardiac death at an early age. In detail, the bile ducts carry bile produced in the liver to the gallbladder, where it is stored, and then to the small intestine. Recent studies have shown that variations in the ABCG5 gene may lead to the production of a sterol transporter protein that pumps more cholesterol into the bile than usual. This causes cholesterol in the bile ducts and gallbladder to exceed the amount of bile fluid that can be dissolved, leading to the formation of gallstones.
