Background

TAGLN2 is a member of the calponin family of actin-bundling proteins that is involved in the regulation of cell morphology, motility, and cell transformation. It is an actin-cross-linking protein containing a calponin homolog (CH) domain, with a MW of 24 kDa. TAGLN2 can regulate various biological processes in cancer cells such as proliferation, differentiation and apoptosis. The dysregulation of TAGLN2 in a variety of malignant tumor types such as colorectal cancer, lung cancer, and breast cancer, has been discovered through proteomic analysis, and thus reveals an important role for TAGLN2 in tumor progression.

The high resolution of liquid chromatography (LC) coupled with fast and sensitive mass spectrometry (MS) methods has helped in the quantitative measurement of TAGLN2 in complex biological specimens, thereby cutting down on the huge expenditure that goes into developing specific immunoreagents.

Creative Biolabs has established comprehensive TAGLN2 IHC assay protocol including optimized block buffer, primary antibody. Signal is generally visualized using standard protocols with HRP conjugated secondary antibody and DAB as the substrate. For negative controls, sections are incubated with normal serum rather than primary antibody. Slides are counterstained with hematoxylin, and representative images will be obtained.

It is reported that TAGLN2 knockdown in glioma cells induced cell cycle arrest at G0-G1 and cell apoptosis, and reduced growth in orthotopic xenografts and can effectively reduce cell proliferation, migration and invasion in bladder cancer cells.

Based on its potentially important role in promoting glioma development, Creative Biolabs provides TAGLN2 silencing services for our clients. Small interfering RNA (siRNA) synthesize service is available. Stable knockdown of TAGLN2 in cells could be generated using lentiviral transduction and knockdown efficiency is evaluated 48 h after transfection by WB.
