Background

ABCG8 is located on chromosome 2p21 and has 13 exons that encode 673 amino acid residues. As a member of the superfamily of ATP-binding cassette (ABC) transporters, ABCG8 plays an important role in the regulation of intestinal plant sterol absorption by excreting plant sterols.

The ABCG5/ABCG8 transporters are functional heterodimers that are co-expressed in the canalicular membrane of the hepatocyte and the apical membrane of the enterocytes. They serve to export cholesterol and phytosterols across both of these membranes, thereby excluding them from circulation. Formation of a heterodimer is an absolute necessity to direct the ABCG5/ ABCG8 heterodimer from the endoplasmic reticulum to the apical membrane. Mutations in either of these half-transporters result in defective biliary sterol transport, leading to the human disease sitosterolemia.

Scientific Validity

Serum cholesterol-standardized campesterol and sitosterol concentrations, as well as their changes after consumption of plant stanol-enriched foods, are related to a variation in the ABCG8 gene that is present in ∼70% of the population. More and more scientists focus on the relationships between changes in serum plant sterol concentrations with ABCG8 polymorphisms after the consumption of plant stanol esters.
