Description
The Jurkat-Tg(Mouse PD-1)-NFAT Luc reporter cell line is a specialized cell line that has been genetically engineered to express a luciferase reporter gene under the control of the nuclear factor of activated T-cells (NFAT) response element. This cell line also expresses the mouse programmed cell death protein 1 (PD-1) receptor, which can be activated by its ligands to regulate T-cell activation and function. This cell line is commonly used in research to study the role of PD-1 signaling in T-cell activation and function, particularly in the context of immune responses and autoimmune diseases

Background
The PD-1 gene, also known as programmed cell death protein 1, is a gene that encodes a protein found on the surface of T cells and other immune cells. PD-1 plays a crucial role in regulating the immune response by inhibiting T cell activity and preventing autoimmunity. However, overexpression of PD-1 can also suppress the immune response against cancer cells, allowing tumors to evade detection and destruction by the immune system. Therapies targeting PD-1, such as checkpoint inhibitors, have been developed to block the interaction between PD-1 and its ligands, releasing the brakes on the immune response and helping the body's immune system to attack and eliminate cancer cells. These treatments have shown significant success in treating various types of cancer, including melanoma, lung cancer, and lymphoma.

Identity testing is required for newly established cell lines. Isoenzyme analysis is used to confirm the identity of the species of a cell line. Alternative methods for identity testing include DNA fingerprinting, STR analysis and karyology.
Virological Safety Testing
A broad range of viruses is susceptible to affecting human cell lines. We can provide in vivo/vitro virus safety assays by utilizing various animal systems. These viruses include: adventitious viruses, bovine viruses, human and simian viruses, porcine viruses, retrovirus and rodent viruses.

Frozen cells should be stored in a liquid nitrogen tank (-150°C~-190°C). After thawing, cells should be immediately transferred to the appropriate culture medium and maintained under recommended culture conditions.
Handling Notes
Frozen cells should be thawed immediately upon receipt and grown according to handling procedure to ensure cell viability and proper assay performance.
Note: Do not freeze the cells upon receipt as it may result in irreversible damage to the cell line.
