Introduction to Mammalian Milk-Based Polyclonal (IgG) Technology

Nowadays, Creative Biolabs has developed a novel polyclonal-mediated technology platform for treating many immune-related diseases. Based on the conventional polyclonal antibody and vaccine discovery process, we cleverly took advantage of the high absorption rate and safety of mammalian milk, especially bovine colostrum, in the human stomach and intestines to design and obtain specific hyperimmune mammalian immunoglobulin G (IgG). After continuous isolation and purification of these highly immune polyclonal IgG (mostly IgG1), a complete portfolio of custom oral drugs or biologics has been developed by our labs.

Features of Mammalian Milk-Based Polyclonal (IgG) Technology

Currently, we can produce this highly specific immunoglobulin against any intestinal pathogen to produce active oral drug precursors or compounds. Bovine IgG, for example, binds to many human pathogenic agents, preventing them from adhering to the intestinal epithelium. In turn, bovine IgG neutralizes experimental human cell infections and limits gastrointestinal inflammation. In addition, studies involving infants and adults have found that bovine IgG or IgG-rich bovine colostrum plays an important role in preventing gastrointestinal tract (GIT) infections and upper respiratory tract (URT) infections. Thus, our technique could be used to block viruses or bacteria on mucosal surfaces, such as GIT, allowing us to develop drugs for various infectious diseases. In addition, this clear dairy source of antibodies makes it easier for our potential drugs to pass most regulatory regimes.

Case Study: Bovine Colostrum Immunoglobulin Target Colonization Factor Antigens (CFAs) in Diarrhea Therapy

In Creative Biolabs, a hyperimmune bovine colostrum antibody target purified CFAs of enterotoxigenic Escherichia coli (ETEC) product has been developed and evaluated for providing a new alternative diarrhea treatment strategy.

In general, several ETEC strains, such as E. coli M424C1 and E. coli H10407, have been used for the preparation and production of CS1, CS3, as well as CFA/I. CFAs were further purified by ammonium sulfate precipitation and used as a bovine vaccine. All dairy cows were vaccinated and administered at a single site. During this time, all potential adverse reactions in the animals were in real-time monitored and no one was observed. Milk from eight different batches (approximately 4-5 weeks) was collected and pooled to produce anti-E. coli bovine milk immunoglobulin. Hyperimmune milk was then processed into a variety of foods, such as milk powder and cheese.

Moreover, anti-CFA, anti-lipopolysaccharide (LPS), and anti-heat-labile toxin (LT) titers were assessed by detecting the binding of milk antibodies to specific antigens by a panel of ELISA assays. The data have shown that these products have a large number of CFA-specific anti-E. coli activity. As a result, antibodies against CFAs and their milk-derived immunoglobulin are an effective method for diarrhea therapy. Our platform technology allows us to develop drugs for a variety of infectious diseases.
