ErbB Receptor Quantification Service

Our ErbB receptor quantification service provides precise measurement and analysis of ErbB family receptors' expression levels in biological samples, aiding in research and diagnostic applications related to cancer and other diseases.

ErbB Receptor

ErbB receptors, also known as epidermal growth factor receptor (EGFR) family receptors, are a group of closely related proteins that play critical roles in cell growth, differentiation, and survival. They are receptor tyrosine kinases that are activated by binding to specific ligands, such as growth factors. The ErbB family consists of four main members:

EGFR (ErbB1): This receptor is widely studied and is involved in the control of cell proliferation, migration, and survival. Aberrant activation of EGFR is associated with various cancers (e.g., lung, breast).

ErbB2 (HER2/neu): This receptor does not have a known ligand and is often overexpressed in certain cancers, particularly breast cancer. It forms heterodimers with other ErbB family members, enhancing their signaling capabilities.

ErbB3 (HER3): This receptor has reduced kinase activity and relies on heterodimerization with other ErbB receptors for effective signaling. It has been implicated in tumor growth and survival.

ErbB4 (HER4): This receptor can have both growth-promoting and growth-inhibiting effects, depending on the context. It can also be involved in development and differentiation processes.

The activation of ErbB receptors typically leads to a cascade of downstream signaling pathways, including the RAS/RAF/MEK/ERK pathway and the PI3K/AKT pathway, which are crucial for promoting cell survival, growth, and differentiation.

ErbB Receptor Quantification Service at Creative Biolabs

Amplified luminescent proximity homotransfer assay is a sensitive assay technology often used to quantify proteins, including their phosphorylated forms. When quantifying phosphorylated (activated) BTK versus total BTK, the assay can leverage specific antibodies that discriminate between the phosphorylated and non-phosphorylated forms of the protein. Creative Biolabs offers custom quantification services. These can be tailored to specific needs using different methodologies.

Other Services

In addition to the above generic ErbB receptor quantification services, Creative Biolabs also provides the following methods and techniques for the assessment of ErbB receptor activation/inactivation:

Downstream Signaling Pathway Analysis
  • The downstream signaling pathways activated by ErbB receptors include the PI3K/AKT, RAS/RAF/MEK/ERK, and PLCγ pathways.
  • Luciferase Reporter Assays: Measure transcriptional activity resulting from pathway activation.
Ligand Binding Assays
  • Assess the binding affinity of ligands (e.g., EGF, NRG) to ErbB receptors, which can indicate receptor activation.
  • Surface Plasmon Resonance (SPR): Measures real-time binding interactions.
  • Radiolabeled Ligand Binding Assays: Use of radioactively labeled ligands to quantify binding.
