Background
Accurate determination of protein concentration is a critical prerequisite for quantitative molecular biology techniques. The bicinchoninic acid methodology relies on the reduction of copper(II) to copper(I) by peptide bonds under alkaline conditions, followed by the highly sensitive, stoichiometric chelation of BCA with the reduced copper to yield a stable chromophore.
