Characteristics of MMP-9
MMP-9 is a member of matrix metalloproteinases (MMPs) which are a family of zinc-dependent endopeptidases with more than 20 different members, such as collagenases, gelatinases, matrilysins, stromelysins and membrane. The gene of human MMP-9 is located at chromosome 20q13.12, which contains 13 exons and 12 introns. Human MMP-9 protein contains the catalytic domain, hemopexin-like domain, signal peptide, propeptide region and the hinge region. The catalytic domain of MMP-9 contains active site, fibronectin type II (FN2) domains and zinc-binding region. This protein contains five calcium ions and two zinc ions.

This review discusses the complex and dual role of MMP9 in cancer and its challenges as a therapeutic target. While it often promotes tumor invasion, metastasis, and angiogenesis, MMP9 can also have tumor-suppressive functions. The article examines its regulation, its wide range of substrates, and its involvement in the tumor microenvironment. It also reviews various strategies to inhibit MMP9, highlighting the difficulties and failures in clinical trials due to toxicity and lack of specificity, and concludes that more selective targeting approaches are needed.

Frequently Asked Questions
Q1: What is the benefit of your functional assays over simple western blotting or qPCR?
A1: While western blotting and qPCR measure protein and gene expression levels, our functional assays directly measure outcomes like cell invasion and migration. This validates whether an observed change in expression translates to a meaningful change in tumor behavior.
Q2: How do you handle assay development for non-standard MMP-9 variants or isoforms?
A2: We understand that MMP-9 can exist in various forms, including pro- and activated forms. Our team can develop custom assays to specifically analyze the activity of these different variants, ensuring that your research is precise and comprehensive.
Q3: Do you offer services to analyze the interaction of MMP-9 with its inhibitors at a molecular level?
A3: In addition to functional assays, we can employ techniques to characterize the binding kinetics and affinity of your compounds to MMP-9. This provides crucial molecular-level data to complement the functional insights from our assays.
