DKK3, also called REIC, belongs to the dickkopf family. It inhibits the canonical Wnt pathway by preventing the interaction of LRP5/6 with Wnt and forming a ternary complex with KREMEN, which promotes the internalization of LRP5/6. DKK3 plays a significant role in vertebrate development, influencing processes like limb and eye formation. In adults, it is implicated in bone formation, cancer, and Alzheimer's disease. The adenovirus carrying REIC/Dkk-3 (Ad-REIC) has been shown to kill tumor cells selectively, though some cancers are resistant to its apoptosis-inducing effects.
