This study presents a novel fourth-generation lentiviral vector platform for in vivo CAR T cell generation. The authors developed CD3- or CD8-targeted vectors using a mixed Nipah envelope with binders, achieving specific and efficient T cell transduction. The SupA2KO-LV genome incorporates the TRiP system to prevent CAR protein packaging. Intravenous injection into humanized mice generated high levels of functional CAR T cells, inducing rapid and sustained B cell aplasia with minimal off-target transduction.
