While many approved and clinical-stage ADCs rely on cytotoxic payloads such as Topoisomerase I inhibitors or microtubule-disrupting agents, the field is now expanding toward more diverse mechanisms of action.

Immune-modulating ADCs represent a growing area of interest, offering the potential to deliver immune-stimulating or immune-suppressive payloads with improved selectivity and biological control. These approaches may help broaden the therapeutic utility of ADC technology beyond conventional cytotoxic strategies and open new opportunities in cancer, autoimmune disorders, and rare diseases.

In this webinar, Dr. Tumey will provide an overview of the history and current clinical status of immune-stimulating and immune-suppressive ADC designs, followed by a discussion of emerging linker technologies for immune-modulating bioconjugates.

The presentation will cover novel linker designs for both immune-stimulating and immune-suppressive conjugates, including legumain-cleavable ADC strategies, self-immolative spacers for the release of alcohol-containing payloads, and the importance of Fc-silencing for immune-modulating ADC platforms.

Dr. L. Nathan Tumey joined Binghamton University in 2017 after 15 years of medicinal chemistry experience in the pharmaceutical and biotechnology industry. His research focuses on the design of antibody-drug conjugates for the treatment of cancer, autoimmune disorders, and rare diseases.

Dr. Tumey is a leading expert in ADC technology, with particular expertise in bioconjugation, linker design, and immune-modulating payloads. His research includes immune-stimulating antibody conjugates, glucocorticoid-antibody conjugates, legumain-cleavable linkers, and site-specific conjugation technologies.

Before joining Binghamton University, Dr. Tumey held key leadership roles in multiple drug discovery programs, including programs focused on obesity, asthma, rheumatoid arthritis, and ADC-based cancer therapeutics. He received his doctorate in chemistry from Duke University under the supervision of Michael Pirrung and continues to advise biotechnology and pharmaceutical companies on ADC-related programs.
