Antibody Reformatting Solutions
Antibody V regions (VH and VL) from a specific antibody can be reformatted into a variety of different fragments including Fab, F(ab)'2, scFv, VHH, minibodies, and also bispecific antibodies combined with a second specificity. Various antibody formats are optimized for tumor or tissue penetration, or expression in production systems. Here are our current available antibody reformatting choices.
Fab

Fab is monovalent and is approximately only a third of the size of a whole IgG;

Fab has good characterized penetration properties into tissues and faster clearance from circulation;

Publication
This publication presents a novel antibody fragment format, FabH3, designed to overcome production limitations of traditional Fab fragments. This new format replaces the constant domains of a Fab molecule with engineered IgG1 CH3 domains that can heterodimerize using an "electrostatic steering" approach. The FabH3 molecule can be produced in the cytoplasm of E. coli using the CyDisCo system (catalyzed disulfide-bond formation system). This method yields a natively folded protein with a comparable binding affinity to the target antigen (SARS-CoV-2 RBD) but at higher soluble yields than its traditional Fab counterpart. The study validates that this format design successfully overcomes the folding and production constraints associated with the CH1 domain found in Fabs, regardless of expression conditions. The results suggest that the replacement of constant domains and the CH3 domain-based heterodimerization do not interfere with the folding and function of the variable domains.
