Background
Lactate dehydrogenase A (LDHA) is a key catalytic subunit of the LDH enzyme complex that plays a crucial role in cellular energy metabolism. It promotes the conversion of pyruvate to lactate while regenerating NAD+ required for continuous glycolysis. Elevated LDHA expression is frequently observed in rapidly proliferating cells and multiple disease states, particularly cancer.
Pathways
LDHA functions in the glycolytic pathway by converting pyruvate to lactate and maintaining intracellular NAD+ levels required for sustained glycolysis. This reaction becomes especially important under hypoxic conditions. LDHA activity is strongly associated with hypoxia-inducible factor (HIF-1α) signaling and the metabolic reprogramming characteristic of tumor cells.
