This review article examines the critical role of tumor-resident memory T (TRM) cells in cancer immunotherapy. Tumor-infiltrating lymphocytes (TILs) contain a substantial proportion of CD8+ TRM cells, which are enriched with tumor-specific T cells and are key players in the anti-tumor immune response. These cells are defined by their expression of integrins (CD103 and CD49a) and the lectin CD69. Despite expressing inhibitory receptors, TRM cells remain highly reactive against cancer cells and exhibit strong cytotoxic activity—especially when PD-1/PD-L1 signaling is blocked. The abundance of CD8+ TRM cells serves as a prognostic marker for patient survival across multiple cancer types and has recently emerged as a predictive biomarker for responses to immunotherapy. The article outlines the defining features of these cells and discusses their potential in developing more effective immunotherapeutic strategies.
