Mammalian cells such as Chinese hamster ovary (CHO) cells have been extensively utilized in the biopharmaceutical industry as the production hosts for recombinant protein therapeutics, especially glycoproteins, due to several key advantages such as scalability, high production rates, and human-like glycosylation. Currently, the majority of therapeutic biologics are manufactured using the CHO cell lines. There are differences in glycosylation between human and nonhuman mammalian cells. In particular, there are two nonhuman glycans, α1,3-Gal and Neu5Gc, that exist in mouse cells. In addition, the glycosylation patterns of proteins expressed in CHO and human cell lines are likely to differ significantly. CHO cells typically do not express GnT-III, resulting in the absence of bisecting GlcNAc residues in their glycoforms. These differences in glycosylation can impact the bioactivity of glycoproteins produced in CHO cells, particularly therapeutic biologics. Therefore, glycoengineering in nonhuman mammalian cells is indispensable for the production of recombinant human glycoproteins.
