    • Preparation of lectin microarrays: We select lectins with affinity and immobilize them on the microarray, such as peanut agglutinin (PNA).
    • Sample processing: We collect serum samples and perform appropriate pre-processing steps to reduce the impact of interfering substances on subsequent analysis.
    • Sample incubation and washing: We add the pre-processed serum samples to the lectin microarray to allow the sugars in them to bind to the lectins. This is followed by a washing step to remove non-specifically bound components.
    • Fluorescent labeling: We then use fluorescently labeled probes to label the sugars that bind to the lectin and form complexes.
    • Data analysis: We use an imaging system to acquire fluorescence signal images on the lectin microarray and perform quantitative analysis on each spot. The results are interpreted using image processing and data analysis software to determine the content and structure of different sugars.

Results: The authors performed a comprehensive glycosylation analysis of canine serum samples by HILIC-UPLC-MS. The results showed that the structural composition and abundance of glycans in dog serum were significantly different from those in human serum. This study provided a reference for serum glycosylation changes in canine disease.
